    以化合物(CAS: 180624-14-0)为原料合成2-炔基-4-甲基苯胺的一般步骤:在室温下,向搅拌的2-乙炔基-4-甲基苯胺(22)(11.32 g,55.6 mmol)的2:1 MeOH/Et2O(250 mL)溶液中加入K2CO3(15.38 g,111 mmol)。在该温度下搅拌1小时后,将反应混合物用水稀释并用CH2Cl2萃取。有机层经MgSO4干燥,过滤并在减压下浓缩,得到2-炔基-4-甲基苯胺(23),为浅棕色固体,无需进一步纯化(6.79 g,93%)。1H NMR(300 MHz,CDCl3)δ 7.14(d,J = 1.5 Hz,1H),6.96(dd,J = 8.2, 1.5 Hz,1H),6.62(d,J = 8.2 Hz,1H),4.11(s,2H),3.36(s,1H),2.21(s,3H)。
