Cis-2-aminocyclohexanol hydrochloride was prepared as follows:
1) To a mixture of cooled m-chloroperbenzoic acid (9.87 g; 57.19 mmol) in 125 ml of CH2Cl2 was slowly added cyclohexene (5.00 g; 51.99 mmol). The reaction mixture was allowed to warm gradually to room temperature overnight. The mixture was filtered, washed with concentrated NaHCO3 and concentrated in vacuum to give a light yellow oily cyclohexene oxide which was used directly in the next step.
2) In a sealed tube, cyclohexene oxide (5.44 g; 48.50 mmol) and ammonium hydroxide solution (10 ml) in 4 ml of methanol were heated to 80°C for 72 hours. After removing the heat, the mixture was concentrated in vacuum to obtain cis-2-aminocyclohexanol hydrochloride.
