Laminin 332 is indispensable for the efficient and specific differentiation of human induced pluripotent stem cells (iPSCs) into corneal epithelial cells. This laminin isoform is a fundamental component of the corneal epithelial basement membrane in vivo, where it provides not just structural anchorage but also essential pro-differentiation signals. When used as a coating substrate in vitro, LN332 mimics this native niche, powerfully directing iPSCs towards the corneal epithelial lineage. It achieves this by engaging with cellular integrins (primarily α6β4), which triggers signaling pathways that suppress alternative cell fates and actively promote the expression of key corneal epithelial markers like KRT3 and KRT12. This makes LN332 a critical tool for generating authentic, functional corneal epithelial cells for applications in disease modeling, drug testing, and the development of regenerative therapies for ocular surface disorders.
