The RB1 antibody is a crucial tool in studying the retinoblastoma protein (pRB), encoded by the RB1 gene, a pivotal tumor suppressor involved in cell cycle regulation. Discovered through research on retinoblastoma, a rare childhood eye cancer, RB1 mutations or deletions disrupt its function, leading to uncontrolled cell proliferation. The pRB protein primarily inhibits G1-S phase progression by binding and sequestering E2F transcription factors, thus blocking DNA replication. Its activity is modulated by phosphorylation, which releases E2F, enabling cell cycle progression. Dysfunctional RB1 is linked to various cancers, including osteosarcoma, lung, and bladder cancers.

RB1 antibodies are widely used in techniques like Western blotting, immunohistochemistry (IHC), and flow cytometry to detect pRB expression, localization, and phosphorylation status in tissues or cell lines. These antibodies aid in cancer research, diagnostics, and therapeutic development, helping assess tumor suppressor status or treatment responses. Specific clones target distinct epitopes or phosphorylation sites (e.g., Ser807/811), requiring careful selection based on experimental needs. Proper sample preparation is critical, as phosphorylation or degradation can affect detection.
