**Background of ANXA5 Antibodies**

ANXA5 (Annexin A5) is a calcium-dependent phospholipid-binding protein belonging to the annexin family, widely expressed in placental, vascular, and immune cells. It plays critical roles in maintaining membrane integrity, inhibiting blood coagulation by shielding phospholipids, and modulating anti-inflammatory and apoptotic processes.

ANXA5 antibodies are autoantibodies targeting this protein, often associated with autoimmune disorders. Notably, they are linked to **antiphospholipid syndrome (APS)**, where their presence may disrupt Annexin A5’s anticoagulant function, increasing thrombosis and pregnancy complication risks. Research suggests these antibodies contribute to placental insufficiency by interfering with trophoblast membrane repair, potentially leading to recurrent miscarriages.

Beyond APS, ANXA5 antibodies are studied in systemic lupus erythematosus (SLE), cardiovascular diseases, and infertility. Their detection aids in diagnosing autoimmune-related thrombotic events or obstetric complications. However, their clinical utility remains debated due to variability in assay standardization and unclear pathogenic mechanisms.

Recent studies explore their role as biomarkers for predicting thrombotic risk or guiding immunotherapy. Understanding ANXA5 antibodies enhances insights into autoimmune pathogenesis and may inform targeted therapeutic strategies, though further validation is needed for routine clinical application.
