CD129. also known as interleukin-9 receptor alpha (IL-9Rα), is a type I transmembrane glycoprotein that belongs to the cytokine receptor superfamily. It pairs with the common gamma chain (γc, CD132) to form the functional receptor for interleukin-9 (IL-9), a pleiotropic cytokine involved in immune regulation. CD129 is primarily expressed on activated T cells, mast cells, eosinophils, and airway epithelial cells. Structurally, it contains a conserved WSXWS motif in its extracellular domain, critical for ligand binding and signal transduction.

IL-9 signaling through CD129 activates JAK-STAT (Janus kinase-signal transducer and activator of transcription) pathways, influencing cell proliferation, survival, and differentiation. It plays roles in Th2-mediated immune responses, allergic inflammation, and asthma pathogenesis. Dysregulated IL-9/CD129 signaling has been implicated in autoimmune diseases, certain cancers (e.g., T-cell lymphomas), and chronic respiratory conditions.

CD129-specific antibodies are essential tools for detecting receptor expression via flow cytometry, immunohistochemistry, or Western blot. They also enable functional studies to block or modulate IL-9 signaling in experimental models, aiding therapeutic research. Recent studies explore CD129 as a potential therapeutic target, with antibodies or inhibitors being developed to mitigate IL-9-driven pathologies. Its restricted expression pattern makes it a candidate biomarker for specific immune disorders.
