The VRK2 (Vaccinia-Related Kinase 2) antibody is a tool used to study the VRK2 protein, a serine/threonine kinase belonging to the VRK family, which shares homology with vaccinia virus B1R kinase. It exists in two isoforms: VRK2A (localized to the endoplasmic reticulum) and VRK2B (mitochondrial), with distinct functional roles. For instance, elevated VRK2 expression is observed in glioblastoma and breast cancer, correlating with poor prognosis, while reduced levels are associated with schizophrenia. The VRK2 antibody enables researchers to investigate its expression, localization, and interaction partners via techniques like Western blotting, immunohistochemistry, and immunofluorescence. Challenges in antibody development include ensuring isoform specificity due to structural similarities between VRK2 isoforms and related kinases. Recent studies highlight VRK2's involvement in autophagy, stress signaling (e.g., ER stress), and Wnt/β-catenin pathways, making it a potential therapeutic target.
