The Brain Expressed X-linked 3 (BEX3) protein, also known as CDX3 or NAG14. is a member of the BEX/GRB7 family involved in neuronal development, apoptosis, and cell cycle regulation. Primarily expressed in the brain, BEX3 interacts with neurotrophic receptors (e.g., TrkA) and signaling pathways like NF-κB, modulating neural differentiation and survival. It acts as an adaptor protein, linking receptor tyrosine kinases to downstream effectors, and influences transcriptional activity by associating with chromatin modifiers. Dysregulation of BEX3 is implicated in neurological disorders and cancers. In neuroblastoma and gliomas, BEX3 exhibits dual roles, acting as a tumor suppressor by promoting apoptosis or as an oncogene by enhancing proliferation, depending on cellular context. Reduced BEX3 expression correlates with poor prognosis in some cancers.

BEX3 antibodies are essential tools for studying its expression patterns, subcellular localization, and interactions. They enable detection in tissues or cell lines via Western blot, immunohistochemistry, and immunofluorescence. Research using BEX3 antibodies has highlighted its potential as a biomarker for neurodevelopmental disorders (e.g., autism) and neurodegenerative diseases like Alzheimer’s. However, functional mechanisms remain partially unresolved, particularly its context-dependent roles in cancer. Commercial BEX3 antibodies vary in specificity, requiring careful validation for experimental accuracy. Ongoing studies aim to clarify its therapeutic potential in targeting BEX3-associated pathways.
