The MACROH2A2 antibody is a research tool used to detect the macroH2A2 histone variant, a member of the macroH2A family (macroH2A1 and macroH2A2). These variants are characterized by a large C-terminal macrodomain appended to a core histone H2A structure. MacroH2A2 is involved in chromatin remodeling and gene regulation, playing roles in X-chromosome inactivation, cellular differentiation, senescence, and tumor suppression. Unlike macroH2A1. which has two splice isoforms, macroH2A2 exists as a single isoform and exhibits distinct expression patterns, often enriched in repressive chromatin regions associated with transcriptional silencing.

Antibodies targeting macroH2A2 enable researchers to study its localization, expression levels, and functional interactions in epigenetic processes. They are critical for techniques like Western blotting, immunofluorescence, and immunohistochemistry, aiding in investigations of its role in diseases, particularly cancer. MacroH2A2 has been implicated in suppressing tumor metastasis and maintaining genomic stability, with reduced expression observed in certain malignancies.
