SUMO4 (Small Ubiquitin-like Modifier 4) is a member of the SUMO protein family, which regulates post-translational modifications by covalently attaching to target proteins. This process, termed sumoylation, influences protein localization, stability, and interactions, thereby modulating cellular processes such as transcriptional regulation, stress responses, and immune signaling. SUMO4 shares structural similarities with other SUMO isoforms (SUMO1-3) but exhibits distinct expression patterns, predominantly in immune tissues like the spleen and kidneys. It has been implicated in autoimmune and inflammatory diseases, notably type 1 diabetes, due to its genetic association with susceptibility loci and its role in regulating NF-κB activity. A SUMO4 polymorphism (M55V) is linked to altered inflammatory responses, though its mechanistic impact remains debated.

SUMO4 antibodies are essential tools for studying its expression, localization, and function. They enable detection of SUMO4-conjugated proteins via techniques like Western blotting, immunoprecipitation, and immunofluorescence. Researchers also use these antibodies to explore SUMO4's involvement in disease pathways, including diabetes, cancer, and neurodegeneration. Challenges in SUMO4 research include cross-reactivity with other SUMO isoforms due to high sequence homology, necessitating careful antibody validation.
