The estrogen-inducible protein pS2. also known as Trefoil Factor 1 (TFF1), is a small secretory protein encoded by the TFF1 gene. Discovered in the 1980s, pS2 was initially identified as a gene transcriptionally activated by estrogen in hormone-responsive breast cancer cells. It belongs to the trefoil factor family, characterized by a conserved three-loop structure critical for mucosal protection and repair in epithelial tissues. pS2 is primarily expressed in the gastrointestinal tract, particularly in gastric mucosa, and plays roles in maintaining mucosal integrity, promoting cell migration, and suppressing inflammation.

Antibodies targeting pS2 are widely used in research to investigate its expression patterns, regulatory mechanisms, and clinical relevance. pS2 overexpression is associated with estrogen receptor (ER)-positive breast cancers, where it serves as a marker of estrogen signaling activity. Its expression is also linked to certain gastric pathologies, including cancer. In diagnostics, pS2 antibodies aid in immunohistochemical (IHC) detection to assess tumor hormone responsiveness or classify subtypes. Studies further explore pS2's dual role as both a tumor suppressor and potential oncogenic factor, depending on context.
