TIM44 (Translocase of the Inner Mitochondrial Membrane 44) is a key component of the mitochondrial protein import machinery, primarily involved in the transport of nuclear-encoded proteins into the mitochondrial matrix. It acts as a peripheral membrane protein anchored to the inner mitochondrial membrane, where it serves as a critical adaptor between the translocase TIM23 complex and the mitochondrial Hsp70 (mtHsp70) chaperone system. TIM44 facilitates the ATP-dependent translocation of precursor proteins by recruiting mtHsp70 to the translocation channel, enabling the binding and folding of incoming polypeptides.

Structurally, TIM44 contains an N-terminal transmembrane domain and a C-terminal acidic domain that interacts with mtHsp70. Its dynamic association with TIM23 and coordination with co-chaperones like Tim14 (DNAJA3) ensure efficient protein import, a process vital for mitochondrial biogenesis and function. Dysregulation of TIM44 has been linked to mitochondrial disorders and cellular stress responses.
