Galectin-8. a member of the galectin family, is a β-galactoside-binding lectin involved in diverse cellular processes, including immune regulation, autophagy, and cell adhesion. It contains two carbohydrate recognition domains (CRDs) with distinct binding preferences, enabling interactions with glycoproteins and glycolipids. Primarily expressed intracellularly, Galectin-8 also localizes to the extracellular matrix, where it modulates cell signaling and pathogen recognition. Its role in autophagy, particularly in targeting damaged lysosomes (a process termed "galectin-directed autophagy"), has drawn significant attention. Additionally, Galectin-8 functions as a danger sensor, promoting inflammation by activating innate immune pathways like the NLRP3 inflammasome.

Antibodies targeting Galectin-8 are critical tools for studying its biological functions and therapeutic potential. They enable detection of Galectin-8 expression in tissues, cell lines, and pathological conditions such as cancer, autoimmune diseases, and infections. Some studies suggest Galectin-8 overexpression correlates with tumor progression, making it a potential biomarker or therapeutic target. Neutralizing antibodies are being explored to inhibit Galectin-8-mediated pathways in inflammatory diseases or to enhance immune responses in cancer immunotherapy. Challenges remain in optimizing specificity due to structural similarities among galectins.
