Coilin antibodies are essential tools in studying Cajal bodies (CBs), membrane-less subnuclear organelles involved in RNA processing and ribonucleoprotein (RNP) complex assembly. Coilin, a 576-amino acid phosphoprotein, serves as a structural and functional scaffold for CBs, though it is not exclusively localized to these structures. First identified in 1991 through autoimmune patient sera, coilin plays roles in small nuclear RNP (snRNP) biogenesis, spliceosomal assembly, telomerase maturation, and transcriptional regulation. Its dynamic behavior—shuttling between nucleus and cytoplasm, forming CBs during G1 phase, and dispersing during stress—makes it a key marker for nuclear organization studies.

Anti-coilin antibodies enable researchers to visualize CBs via immunofluorescence, detect coilin expression by Western blot, or isolate CB-associated complexes through immunoprecipitation. They are widely used to investigate CB dynamics in cell differentiation, viral infection, stress responses, and diseases like spinal muscular atrophy or cancer. Commercially available antibodies target specific epitopes (e.g., N-terminal for Santa Cruz Biotechnology, C-terminal for Abcam), with validation including colocalization with CB markers (SMN, fibrillarin) and knockout cell line testing. Recent studies also explore coilin's non-CB functions in DNA repair and microRNA processing, expanding the antibody's research applications. Proper controls remain critical due to coilin's nucleolar localization in some contexts and antibody cross-reactivity risks.
