Kallikrein 5 (KLK5) is a serine protease belonging to the tissue kallikrein family, encoded by the *KLK5* gene. It plays a critical role in skin homeostasis, particularly in desquamation (shedding of dead skin cells) and barrier function. KLK5 cleaves cell adhesion proteins like desmoglein-1 and corneodesmosin, regulating epidermal shedding. Dysregulation of KLK5 activity is linked to skin disorders such as Netherton syndrome, atopic dermatitis, and psoriasis, where excessive proteolysis disrupts skin integrity. In Netherton syndrome, mutations in the *SPINK5* gene lead to a deficiency of LEKTI, a protease inhibitor that normally regulates KLK5. resulting in uncontrolled KLK5 activity and severe skin inflammation.

KLK5 antibodies are valuable tools for research and diagnostics. They enable detection of KLK5 expression levels, localization, and activity in tissues, aiding studies on its pathological roles. Therapeutic monoclonal antibodies targeting KLK5 are under exploration to inhibit its overactivity in inflammatory skin diseases. Additionally, KLK5 antibodies help elucidate its interactions with other proteases and inhibitors in proteolytic cascades.
