描述	Otaplimastat HCl is a matrix metalloproteinase (MMP) inhibitor that blocks NMDA receptor-mediated excitotoxicity competitively and exhibits antioxidant activity,In oxygen-glucose-deprived endothelial cells, otaplimastat suppressed the activity rather than protein expression of MMPs by restoring the level of tissue inhibitor of metalloproteinase (TIMP) suppressed in ischemia, and consequently reduced vascular permeation. serving as a tool for research into brain ischemic injury and neuroprotection.
体外活性	Otaplimastat HCl (87.5-350 μM，20 min) 可通过竞争性作用保护神经元细胞，使其免受 NNMDA诱导的细胞死亡 [1]。 Otaplimastat HCl (350 μM) 能够抑制原代培养神经元中 NMDA受体激活后引发的钙离子（Ca²⁺）内流 [1]。 Otaplimastat HCl (2-200 μM) 预处理 4 小时时，显著抑制过氧化氢（H₂O₂）诱导的细胞死亡，并减少活性氧ROS的生成 [2]。
体内活性	在大脑中动脉闭塞（MCAO）模型中，Otaplimastat HCl (10-20 mg/kg) 腹腔注射（闭塞前 30 分钟及再灌注后 1 小时各给药一次）能够防止缺血性神经元死亡 [1]。 Otaplimastat HCl (5-10 mg/kg) 每日进行腹腔注射，连续给药 10 天后，可减轻脑卒中引起的运动功能损伤 [2]。
