How effective is ammonium sulphate for purifying antibodies?
The ammonium sulphate precipitation method is a long-established and straightforward technique for purifying antibodies from serum and egg yolk. However, this method can compromise antibody purity to some extent. Studies have shown that the combined treatment of ammonium sulphate and octanoic acid improves both the yield and purity of antibodies in goat serum. In contrast, when egg yolk is treated with ammonium sulphate alone, although the yield is higher, the purity of the antibodies is lower. Treatment of goat serum using the combined ammonium sulphate-octanoic acid precipitation method yields 6.67 mg/mL of purified antibody, whereas treatment with ammonium sulphate alone yields only 4.49 mg/mL. However, in the case of IgY, the combined ammonium sulphate-octanoyl acid precipitation method yielded only 8.90 mg/mL of purified antibody, whereas treatment with ammonium sulphate alone yielded 13.80 mg/mL of purified antibody.
