What conditions has rapamycin been approved by the FDA to treat?

Rapamycin is an antibiotic discovered in the 1970s from Streptomyces hygroscopicus on Easter Island (Rapanui). It has been approved by the US Food and Drug Administration (FDA) for the prevention of transplant rejection and for the treatment of tuberous sclerosis (TSC; caused by mutations in the TSC1 or TSC2 genes) as an antiepileptic therapy. Rapamycin delays cellular ageing, improves healthspan and extends lifespan in animal models by inhibiting the mTOR pathway. However, due to its potential anti-ageing benefits, the drug is frequently used for off-label purposes.
