What is the catalytic principle of DMAP?

DMAP, the full name of 4‑Dimethylaminopyridine, is a highly active pyridine organic base and nucleophilic catalyst. It is mostly white or off-white crystalline powder at room temperature. It is often used to efficiently catalyze acylation, esterification and other reactions in organic synthesis. The dimethylamino group on the pyridine ring in the DMAP molecule has a strong electron-donating effect, which significantly increases the electron cloud density and nucleophilicity of the pyridine nitrogen atom. It first forms a highly active N-acylpyridinium salt intermediate with the acylating reagent. The reactivity of this intermediate is much higher than that of the original acylating reagent, and it can quickly react with nucleophilic substrates such as alcohols and phenols. At the same time, DMAP is regenerated and continues to participate in the next round of catalysis, thereby greatly accelerating the reaction rate and reducing reaction conditions.
