Diisopropyl azodicarboxylate, abbreviated as DIAD, is an orange-red oily liquid commonly used as an important azo organic reagent in fine chemical and pharmaceutical synthesis. In the Mitsunobu reaction, it synergistically interacts with triphenylphosphine to form an active intermediate through a redox process. This intermediate activates the hydroxyl group of the alcohol and promotes nucleophilic substitution reactions, while simultaneously achieving configuration inversion of chiral alcohols. It not only provides a crucial electron transfer carrier for the reaction but also efficiently binds reaction byproducts, driving the reaction forward.
